Protocol for making x 10 times concentrated YNB (Yeast Nitrogen Base) solution

The final concentration of YNB in media depends on the type of YNB

ComponentC (g/L)Ammonium sulfateAmino acids
YNB w/o amino acids w ammonium sulfate6.7
YNB6.7
YNB w/o ammonium sulfate1.7

YNB has:

  • L-Histidine HCl
  • DL-Methionine
  • DL-Tryptophan

YNB w ammonium sulfate has 5 g/L ammonium sulfate.

10 x YNB w ammonium sulfate w amino acids

Dissolve 13.4 g YNB w (NH4)2SO4 in 200 mL water. Sterilize by autoclaving.

10 x YNB w/o ammonium sulfate

Dissolve 13.4 g YNB w/o (NH4)2SO4 in 200 mL water. Sterilize by autoclaving.
