Protocol for making x 10 times concentrated YNB (Yeast Nitrogen Base) solution
The final concentration of YNB in media depends on the type of YNB
| Component | C (g/L) | Ammonium sulfate | Amino acids |
|---|---|---|---|
| YNB w/o amino acids w ammonium sulfate | 6.7 | ✅ | ✅ |
| YNB | 6.7 | ✅ | ❌ |
| YNB w/o ammonium sulfate | 1.7 | ❌ | ❌ |
YNB has:
- L-Histidine HCl
- DL-Methionine
- DL-Tryptophan
YNB w ammonium sulfate has 5 g/L ammonium sulfate.
10 x YNB w ammonium sulfate w amino acids
Dissolve 13.4 g YNB w (NH4)2SO4 in 200 mL water. Sterilize by autoclaving.
10 x YNB w/o ammonium sulfate
Dissolve 13.4 g YNB w/o (NH4)2SO4 in 200 mL water. Sterilize by autoclaving.
